Rockland Immunochemicals has collaborated with Bio-Rad to develop an enhanced multiplex fluorescent western blotting protocol. The assays produce high resolution immunoblots on PVDF membrane, providing increased visibility and profiling of low abundance proteins.
Rockland DyLight™ fluorochrome conjugated secondary antibodies are used to probe the membrane and the Molecular Imager® VersaDoc™ MP 4000 and the ChemiDoc™ XRS+ imaging system developed by Bio-Rad Laboratories, provides clear, accurate results. Several targets can be detected and imaged simultaneously, resulting in blots that show enhanced detection of distinct target bands labeled by a unique identifying color. Simultaneous detection helps cut time, while ensuring the target epitopes are still easily identified.
Rockland also collaborated with Bio-Rad on a two-dimensional workflow validation to develop an improved standardized immunoassay for HCP detection. This collaboration resulted in a method using a robust 2D SDS-PAGE using the PROTEAN® i12™ IEF System, ReadyStrip™ IPG Strips, and western blotting assay to validate the efficacy of immunocoverage for novel anti-HCP polyclonal antibodies. Validation is performed using BioRad’s PDQuest 2-D Analysis Software.
Rockland offers KCAJ07, an E.coli HCP Western Blot kit that contains two-dimensional validated antibodies, HRP conjugated secondary antibodies, blocking and wash solutions and E.coli control proteins. Rockland’s two-dimensional validated antibodies have been shown to react with more than 300 E.coli proteins from SDS/DTT solubilized E.coli cells.
