Background

Proinflammatory cytokine tumor necrosis factor-α (TNF-α) contributes to a variety of inflammatory responses and programmed cell death. Notch receptor and its ligand participate in cell fate decisions during vertebrate development and are associated with several human disorders, including a T-cell lymphoma. TNF-α, notch, and its ligand delta, are all membrane-bound molecules, which are cleaved by proteases to release mature proteins or functional receptor.  ADAM10, a metalloprotease-disintegrin in the family of mammalian ADAM (for a disintegrin and metalloprotease), was recently identified to cleave TNF-α, notch and its ligand delta.  The genes encoding human, mouse, and bovine ADAM10 were recently cloned and designated ADAM 10, kuzbanian (KUZ) and MADM, respectively.  ADAM10 mRNA is expressed in a variety of human and bovine tissues. Anti-ADAM10 is ideal for investigators involved in NF-kappaB, Apoptosis, Cytokins and Growth factor research.

This IgG fraction antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to a region near the carboxy terminus of human ADAM10 protein.
ADAM10 is Protein A purified.  The product is affinity chromatography purified via peptide column.  A BLAST analysis was used to suggest cross-reactivity with ADAM10 protein from human, bovine and rat based on 100% homology with the immunizing sequence.  Partial reactivity is expected against ADAM10 from mouse based on a single amino acid change relative to this sequence.  Reactivity against homologues from other sources is not known.

Application Details

ADAM10 antibody has been tested for use in ELISA, immunohistochemistry and western blotting.  Specific conditions for reactivity should be optimized by the end user.  Expect a band approximately 84 kDa in size corresponding to ADAM10 protein by western blotting in the appropriate cell lysate or extract.

Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.
